چکیده
Pelleting inactivates SODle viruses and aggregates other dispersed :material. Methods for separation should be developed in which desired particles are always kept in suspension. Rotors that cODlbine continuous-flow centrifugation with isop"ycnic banding can achieve this effect. In our SySteDl a liquid density gradient is set up in a spinning rotor next to the wall in each of 4 sector-shaped cODlpartDlents. The virus-containing suspension is made to flow over the centripetal surface of the gradient between the gradient and the tapered core of the rotor. Virus particles sedi:ment out of the streaDl into the iDlprisoned gradient. At the co:mpletion of a run, co:mponents of the gradient are recovered by displacing the gradient centripetally with a dense fluid and channeling the gradient out a center line.-Nat Cancer Inst Monogr 21: 199-216, 1966.

Purification of large quantities of influenza virus by density gradient centrifugation.
New zonal centrifuges can conveniently process as much as five orders of magnitude (10(5)) greater sample volumes than conventional swinging-bucket rotors. The continuous-sample-flow-with-banding versions may be used in series with ancillary purification procedures. Here we have studied the combined process: absorption and elution of influenza virus with barium sulfate followed by concentration...
متن کاملAn introduction to particle separations in zonal centrifuges.
Zonal centrifugation, based either on sediInentation rate or on isopycnic banding in liquid density gradients, is useful for high-resolution separation of subcellular particles. However, the amoun t of material which may be separated is extremely limited when conventional centrifuges are employed. To increase the capacity and, in certain instances, the resolution of the method, the possibility ...
متن کاملDensity Gradient Centrifugation in Angle-head Rotors.
Preparative isopycnic density-gradient centrifugation is ordinarily done in high-speed swinging-bucket rotors such as the Spinco SW 39 and SW 25. In addition to requiring this special equipment, the technique is severely limited by the small number of samples and the low total-sample volume which may be used. In recent studies on methods for isolating small numbers of virus particles from tissu...
متن کاملIsolation of native glycogen by combined rate-zonal and isopycnic centrifugation.
Rat liver glycogen was separated into fractions of uniform particle size by the rate-zonal centrifugation of homogenates through cesium chloride and sucrose density gradients. Glycogen was then isolated from other cell constituents having similar sedimentation coefficients by isopycnic-zonal centrifugation in CsCI density gradients to effect a density separation. A density of 1.62 to 1.65 was i...
متن کاملAustralia antigen: large-scale purification from human serum and biochemical studies of its proteins.
Biophysical techniques are described for the large-scale isolation of Australia antigen (Au) from unit quantities of human serum by using the batch-type zonal centrifuge rotors. A three-step procedure involving isopycnic banding of the particle in CsCl density gradients and rate-zonal centrifugation on sucrose gradients resulted in a highly purified Au preparation which was used for biochemical...
